一般描述
脂被蛋白是白色和褐色脂肪细胞内用于贮存中性脂质的脂滴的表面蛋白。肾上腺皮质、卵巢中的类固醇生成细胞和睾丸间质细胞存在内含胆固醇酯的较小脂滴,其表面也包被少量的脂被蛋白。脂滴包被蛋白具有多种同工型,这是由于差异剪接事件引起的。脂滴包被蛋白A在脂肪细胞和类固醇生成细胞中含量最高。脂滴包被蛋白B是脂肪细胞中的次要形式。类固醇生成细胞选择性表达脂滴包被蛋白C和D。
免疫原
与人脂滴包被蛋白A的氨基酸残基492-505相对应的合成肽,其中C-末端添加了与KLH偶联的半胱氨酸。相应的序列在小鼠和大鼠中相差一个残基。
生化/生理作用
脂滴包被蛋白可作为分子开关,通过保护甘油三酯防止其水解而抑制基础的脂肪分解,参与调控脂肪细胞的甘油三酯贮存。脂滴包被蛋白还通过激素敏感脂肪酶(HSL)和非HSL来增强环磷酸腺苷(cAMP)依赖蛋白激酶(PKA)激活的脂肪分解。脂被蛋白敲除小鼠的脂肪组织减少,并且能抵制饮食导致的肥胖。它们的脂质贮存脂滴包被未被PKA磷酸化的脂肪分化相关蛋白(ADRP,adipophilin)。
