Biochem/physiol Actions

蛋白质磷酸化和脱磷酸化是改变真核细胞中蛋白质功能的基本信号传导机制。在正常组织中，磷酸化是一种罕见的遗传翻译后活动，但是磷酸化细胞蛋白质的丰度在多个激活过程后增加了数倍。被磷酸化的主要氨基酸是酪氨酸、丝氨酸或苏氨酸（pTyr/pSer/pThr），且每种氨基酸都有对其磷酸化的特异性激酶和对其脱磷酸化的特异性磷酸酶。针对于磷酸化残基的单克隆和多克隆抗体已经生成，并可用于分析和制备工具进行磷酸化细胞蛋白的鉴定、定量和免疫亲和分离。

通过 ELISA 和斑点印迹，该抗体以游离氨基酸形式或与载体（如 BSA 或 KLH）的缀合，与磷酸化丝氨酸特异性反应。与非磷酸化丝氨酸、磷酸苏氨酸、磷酸酪氨酸、AMP 或 ATP 无交叉反应。该抗体已用在免疫印迹中定位一些含磷酸丝氨酸蛋白质。由于识别位点的空间位阻，该抗体可能无法识别已知含有磷酸化丝氨酸的某些蛋白质。
