Biochem/physiol Actions

Rab GTPases是真核细胞不同亚细胞区室间膜运输的中心调节因子。Rab GTPases的调节能力取决于其在与结合的不活跃状态和与GTP结合的活跃状态之间循环的能力。Rab蛋白的激活再加上其与细胞膜的结合，允许其招集下游效应蛋白到亚细胞区室的细胞质表面。通过其效应蛋白，Rab GTPases可调节囊泡形成、肌动蛋白和微管蛋白依赖性囊泡运动，以及膜融合。Rab7调节从早期到晚期核内体以及从晚期核内体到溶酶体的囊泡运输。Rab7还参与了晚期自噬液泡的成熟。Rab7效应物包括Rab相互作用溶酶体蛋白(RILP)——通过介导动力蛋白/动力蛋白运动的招集来控制晚期核内体和溶酶体运输、Rab-7相互作用的环指蛋白(Rabring7)和hVPS34/p150复合物。

General description

Rab7是小鸟苷三磷酸酶(GTPase)的Rab族成员。该族属于小鸟苷三磷酸酶大鼠肉瘤(Ras)的超级家族。Rab蛋白包括鸟嘌呤核苷酸结合的保守区域和具有半胱氨酸基序的高变COOH-端域，与亚细胞靶向有关。带一个或两个香叶酰香叶酰组的半胱氨酸基序的翻译后修饰对于膜结合和Rab蛋白的正确细胞内定位至关重要。人类染色体中的基因Rab7a定位于3q21.3。
