General description

单克隆抗AGO2（大鼠IgG2a同种型）来自杂交瘤11A9 ，该杂交瘤是通过小鼠骨髓瘤细胞（P3X63Ag8.653）和以对应于人AGO2 1-15氨基酸的肽免疫的大鼠脾细胞融合产生。单克隆抗AGO2 抗体可识别人、猴、牛、大鼠和小鼠AGO2。Argonaute 2（Ago2）由定位于人染色体8q24.3的基因编码。编码的蛋白属于Ago蛋白家族。 AGO2主要位于分散的细胞质和应激颗粒。

Biochem/physiol Actions

Argonaute 2（Ago2）在短干扰RNA介导的基因沉默中起关键作用。在人乳腺癌细胞系中，编码的蛋白在转录和翻译后水平上进行调节。Ago2和micro-RNA活性增强与乳腺癌细胞系的肿瘤进展有关。已经在肝细胞癌（HCC）和头颈鳞状细胞癌患者中观察到该基因的过度表达。

Physical form

溶于含有15 mM叠氮化钠的0.01M磷酸盐缓冲液（pH 7.4）。

Preparation Note

储存温度-20°C。为长期使用，可将本品在2-8°C下储存长达一个月。对于长期储存，将工作分装样品冷冻在-20℃下。不建议重复冷冻和解冻，或储存在无霜冰箱中。如果长期储存时出现轻微混浊，使用前通过离心澄清溶液。如果在12小时内未使用，应丢弃工作稀释样品。

Other Notes

在上样到SDS-PAGE上之前，不应煮沸细胞裂解物，而应该在60℃加热15分钟。为了使用各种技术和制剂获得最佳结果，我们建议通过滴定确定最佳工作稀释度。
