General description

β-微管蛋白属于微管蛋白亚家族，微管蛋白是微管的主要合成砌块。β-微管蛋白的分子量为55kDa。β−微管蛋白的结构特征在于具有被a--螺旋包围的两个β-折叠的核心。它还含有一个具有鸟嘌呤核苷酸结合区的N-末端结构域，一个具有紫杉醇结合位点的中间结构域，以及一个含有分子马达蛋白结合表面的C-末端结构域。人β-微管蛋白由七种亚型组成（β I （I类)、 β II (II类)、 β III （III类）、 β IVa （IVa类）、β IVb （IVb类）、β V （V类）和β VI （VI类）。

单克隆抗-β-微管蛋白(鼠IgG1同种型)来源于D66 杂交瘤，该杂交瘤是由小鼠骨髓瘤细胞和海胆（Lytechinus pictus）精子轴丝蛋白免疫的BALB/c小鼠脾脏细胞融合产生。

Application

Biochem/physiol Actions

可识别所有小鼠大脑β-微管蛋白同种型（β和β′亚群）和海胆β2-微管蛋白。抗体识别的表位位于β-微管蛋白的C末端区域（海胆β2-微管蛋白中的氨基酸427-432）。

基因突变可导致各种神经元迁移障碍，如无脑回、巨脑回和多小脑回畸形。B2702肽能够与β- 微管蛋白结合并抑制自然杀伤（NK）细胞的细胞毒性，它影响微管聚合，从而破坏细胞骨架组织和微管蛋白的分子伴侣活性。β-1微管蛋白也称为VI类或TUBB1，在血小板生成中具有至关重要的作用，被认为是免疫性血小板减少症（ITP）的潜在候选生物标志物。
