一般描述

单克隆抗黏着斑蛋白（小鼠 IgG1 亚型）抗体来自于免疫小鼠骨髓瘤细胞和脾细胞相互融合而产生的杂交瘤细胞。
黏着斑蛋白（VCL）基因含有22个外显子，位于人类染色体17q21.2。该基因编码存在于粘附型细胞连接中的117kDa的细胞骨架蛋白。VCL是一种普遍表达的细胞粘附蛋白。该蛋白有一个球状头部和一个尾部结构域，两者通过富含脯氨酸的短序列相互连接，头部与尾部之间的分子内相互作用掩盖了蛋白中的多个配体结合位点。

特异性

抗体与培养的鸡成纤维细胞反应最好。也可以用牛、人或小鼠细胞获得标签。

免疫原

来自鸡肫的平滑肌肉蛋白。

应用

小鼠抗-Vinculin 单克隆抗体可用于：
  • 蛋白质印迹。

生化/生理作用

黏着斑蛋白可能是肌动蛋白纤维束末端和质膜之间的连接子。有人提出黏着斑蛋白可能与肌动蛋白束的跨膜诱导有关。
黏着斑蛋白（VCL）是一种细胞骨架蛋白，在调控细胞间连接和细胞基质连接中发挥重要作用。VCL促进正常的胚胎发育和心脏功能。该基因的异常或突变会导致心肌病。VCL在细胞迁移和形态发生过程中也起着至关重要的作用。VCL可促使整合素和钙粘蛋白与肌动蛋白细胞骨架结合。黏着斑蛋白在整合素αIIbβ3的信号传导和功能中发挥重要作用，这是血小板聚集所必需的。
