General description
Muscleblind-like protein 1 (UniProt: Q9NR56; also known as Triplet-expansion RNA-binding protein) is encoded by the MBNL1 (also known as EXP, KIAA0428, MBNL) gene (Gene ID: 4154) in human. MBNL1 is an RNA splicing protein that mediates pre-mRNA alternative splicing regulation. It can act either as activator or repressor of splicing on specific pre-mRNA targets. It is highly expressed in cardiac and skeletal muscle and during myoblast differentiation and is weakly expressed in other tissues. It is shown to inhibit cardiac troponin-T (TNNT2) pre-mRNA exon inclusion but induces insulin receptor (IR) pre-mRNA exon inclusion in muscle. It regulates the TNNT2 exon 5 skipping through competition with U2 auxiliary factor 2 (U2AF2). MBNL1 binds to the stem-loop structure within the polypyrimidine tract of TNNT2 intron 4 during spliceosome assembly and inhibits the formation of spliceosome A. MBNL1 gene is one of the most consistently overexpressed genes in MLL-rearranged leukemia and its loss significantly impairs propagation of murine and human MLL-rearranged leukemia in vitro and in vivo. Mutations in MBNL1 gene have been linked to dystrophia myotonica that is characterized by myotonia and muscle wasting in the distal extremities. Seven different isoforms of MBNL1 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Biochem/physiol Actions
Clone 4A8 is a ZooMAb® mouse recombinant monoclonal antibody that specifically detects Muscleblind-like protein 1 (MBNL1).
Physical form
Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
