General description
Glutamate decarboxylase 1 (UniProt: Q99259; also known as EC:4.1.1.15, 67 kDa glutamic acid decarboxylase, GAD-67, Glutamate decarboxylase 67 kDa isoform) is encoded by the GAD1 (also known as GAD, GAD67) gene (Gene ID:2571) in human. GAD67 is responsible for the conversion of glutamic acid to gamma-aminobutyric acid (GABA), the major inhibitory transmitter in higher brain regions, and putative paracrine hormone in pancreatic islets. Two molecular forms of GAD have been described (65 kDa and 67 kDa) that display 64% amino acid homology and both forms are expressed in the CNS, pancreatic islet cells, testis, oviduct, and ovary. Between the two isoforms, GAD67 is responsible for over 90% of basal GABA synthesis and is produced at limiting levels in the brain. These isoforms are regionally distributed in cytoplasm in the brains of rats and mice. GAD67 expression is shown to change during neural development in rat spinal cord and its expression seems to be particularly plastic and can change in response to experimental manipulation and disease progression. Mutations in GAD1 gene are known to cause spastic quadriplegic cerebral palsy, a non-progressive disorder of movement and/or posture resulting from defects in the developing central nervous system. Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
