Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected TCF25 in HeLa cell lysate.
Tested applications
Western Blotting Analysis: A representative lot detected TCF25 in lysates from HEK293 cells (1:10,000 dilution) and A431cells (1:1,000 dilution) and recombinant Human TCF25 (1:1,000 dilution).
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TCF25 in human kidney tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TCF25 in HeLa and NIH3T3 cells.
Enzyme Immunoassay Analysis (ELISA): A serial of dilutions of this antibody detected full-length, recombinant Human TCF25.
Affinity Binding Assay: A representative lot of this antibody bound full-length, recombinant Human TCF25 protein with a KD of 2.2 x 10-7 in an affinity binding assay.

Transcription factor 25 (UniProt: Q9BQ70; also known as TCF-25, Nuclear localized protein 1) is encoded by the TCF25 (also known as KIAA1049, NULP1) gene (Gene ID: 22980) in human. TCF25 is a member of the basic helix-loop-helix (bHLH) family of transcription factors that are important in embryonic development. It is mainly localized in the nucleus but may also be present in cytoplasm to some extent. In the embryo, its highest levels are expressed in the brain and in the adult, highest expression is observed in the heart. It acts as a transcriptional repressor and is reported to repress transcription of serum response factor (SRF) in vitro and may play a role in cardiac development. Its expression is consistently reduced in the failing hearts of patients and in hypertrophic mouse hearts. Overexpression of TCF25 is shown to induce cell death in osteosarcoma Saos2 cells with DNA fragmentation. It renders cancer cells more prone to cell death following treatment with chemotherapeutic drugs, such as staurosporine. It is reported to bind to XIAP and this interaction increases apoptotic cell death. 外形
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
