General description
Metalloproteinase inhibitor 1 (UniProt: P01033; also known as Erythroid-potentiating activity, EPA, Fibroblast collagenase inhibitor, Collagenase inhibitor, Tissue inhibitor of metalloproteinases 1, TIMP-1) is encoded by the TIMP1 (also known as CLGI, TIMP) gene (Gene ID: 7076) in human. The tissue inhibitors of metalloproteinases (TIMPs) represent a family of ubiquitous proteins which are natural inhibitors of the matrix metalloproteinases (MMPs). There are four known members of the TIMP family that are designated as TIMP-1, 2, 3, and 4. TIMPs are released during tissue damage to facilitate tissue remodeling through degradation and reorganization of the extracellular matrix. During this process MMPs are involved in inflammatory response through proteolytic maturation of cytokines. These activities are regulated through a 1:1 stoichiometric interaction of MMPs with TIMPs and these interactions are tightly controlled. TIMP-1 is synthesized with a signal peptide (aa 1-23), which is subsequently cleaved off to produce the mature protein (aa 24-207). It is the best characterized inhibitor of MMP activity and regulates 14 of the 24 known MMPs. It inhibits MMP activity through the binding of its N-terminus with the targeted MMP, which results in chelation of Zn2+ from the enzyme active site. TIMP-1 levels are elevated in plasma of cancer subjects and its higher levels are associated with worse clinical outcomes. Stroma of human prostate and colon cancer is reported to express higher levels of TIMP-1 compared to their normal counterparts. This increased expression is reported to promotes tumor growth. Biochem/physiol Actions
Clone 1H19 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects TIMP-1. It targets an epitope within 15 amino acids from the N-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
