Mothers against decapentaplegic homolog 7 (UniProt: O15105; also known as MAD homolog 7, Mothers against DPP homolog 7, Mothers against decapentaplegic homolog 8, MAD homolog 8, Mothers against DPP homolog 8, SMAD family member 7, SMAD 7, Smad7, hSMAD7) is encoded by the SMAD7 (also known as MADH7, MADH8) gene (Gene ID: 4092) in human. At least nine SMAD proteins have been described in vertebrates that are classified as: (a) receptor-activated SMADs (R-SMADs), (b) co-mediator SMADs, and (c) inhibitory SMADs (I-SMADs). SMAD6 and 7 belong to the group of I-SMADs. SMAD7 is ubiquitously expressed protein with higher expression observed in the lung and vascular endothelium. It is a key negative regulator of TGF-β signaling and plays an important role in modulating a large array of biological processes. Quality Control Testing

Evaluated by Western Blotting in SH-SY6Y cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SMAD7 in SH-SY5Y cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected SMAD7 in A549 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SMAD7 in U-251 MG cells.

Affinity Binding Assay: A representative lot of this antibody bound SMAD7 with a KD of 1.2 x 10^-6 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected SMAD7 in Human kidney tissue sections.

Clone 2D15 is a ZooMAb^® rabbit recombinant monoclonal antibody that specifically detects SMAD7. It targets an epitope within 24 amino acids from the C-terminal region.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
