DNA polymerase theta (UniProt: O75417; also known as EC:2.7.7.7, DNA polymerase eta, POLQ) is encoded by the POLQ (also known as POLH) gene (Gene ID: 10721) in human. POLQ is a homomultimeric enzyme that is minimally expressed in normal tissues but is overexpressed in tumors. Its expression is observed in testis tissue. It contains a helicase-like domain at its N-terminus and a polymerase domain at its C-terminus. They are separated by a large unstructured central domain. It promotes microhomology-mediated end-joining (MMEJ), an alternative non-homologous end-joining (NHEJ) machinery that is triggered in response to double-strand breaks in DNA. It acts by binding directly the 2 ends of resected double-strand breaks, allowing micro homologous sequences in the overhangs to form base pairs. It then extends each strand from the base-paired region using the opposing overhang as a template. It requires partially resected DNA containing 2 to 6 base pairs of microhomology to perform MMEJ. Clone 2F5 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects DNA polymerase theta (POLQ). It targets an epitope within 23 amino acids from the N-terminal half.

Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
