General description
ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb® antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. E3 SUMO-protein ligase RanBP2 (UniProt: P49792; also known as EC:2.3.2.-, 358 kDa nucleoporin, Nuclear pore complex protein Nup358, Nucleoporin Nup358, Ran-binding protein 2, RanBP2, p270) is encoded by the RANBP2 (also known as NUP358) gene (Gene ID: 5903) in human. It is a multifaceted protein that plays a crucial role in nucleocytoplasmic transport, RNA processing and is an important component of the nuclear export pathway. Structurally, RanBP2 is characterized by a large size, comprising approximately 3,224 amino acids, and contains multiple repeats (TPR1-7), FG repeats and multiple Ran-binding domains. RanBP2 has been shown to be involved in the process of sumoylation, a post-translational modification that affects protein function, localization, and stability. RanBP2 is expressed in a wide range of human tissues, including high levels in the brain, liver, and neuronal cells. The protein is predominantly localized in the nuclear pore complexes (NPCs), where it functions as a docking site for import and export factors involved in the transport of proteins and RNA between the nucleus and cytoplasm. FG repeats are interaction sites for karyopherins (importins/exportins) and form an affinity gradient, guiding the transport proteins unidirectionally with their cargo through the NPC. RanBP2 also participates in various cellular processes, including the regulation of gene expression and the assembly of ribonucleoprotein complexes. RANBP2 plays a complex and multifaceted role in cancer, acting as both a potential tumor suppressor and an oncogenic factor depending on the context. A research study reported that loss of RanBP2 or disruption of its RBD2/RBD3 domains in retinal pigment epithelium reproduces degeneration resembling neovascular AMD, linked to altered ubiquitin-proteasome pathways and Mmp11 activity. RANBP2 mutations can cause Acute Necrotizing Encephalopathy (ANE1), a severe neurological disease triggered by viral infections, leading to seizures and coma. Recent studies underscore the potential of RanBP2 as a therapeutic target in cancer treatment and neurodegenerative diseases. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Biochem/physiol Actions
Clone 1B8 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects RanBP2. It targets an epitope within 25 amino acids from the Internal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
