General description
Integrin beta-2 (UniProt: P05107), also known as CD18, Complement receptor C3 subunit beta and Cell surface adhesion glycoproteins LFA-1/CR3/p150,95 subunit beta. It is encoded by the ITGB2 gene (Gene ID: 3689) in human. Integrin β2 (CD18) is a type I transmembrane protein that forms a heterodimer with a variable chain (CD11a-d) to create different integrin complexes. It consists of a large extracellular domain responsible for ligand binding, a single transmembrane helix, and a short cytoplasmic tail that interacts with intracellular signaling molecules. Integrin β2 is primarily expressed on leukocytes, including monocytes, and lymphocytes with highest expression in neutrophils. Subsets of T cells, B cells, and natural killer cells. Expression levels can vary depending on cell type, activation state, and tissue distribution and can be induced by inflammatory cytokines and immune cell activation. It plays a role in signaling pathways that regulate cell activation, differentiation, and immune responses, mediates cell adhesion and migration by binding to ligands such as intercellular adhesion molecules (ICAMs) and iC3b fragment of the third complement component. Through these interactions, it is involved in processes like immune cell recruitment, extravasation, and phagocytosis. Mutations or deficiencies in integrin beta-2 have been linked to a rare autosomal recessive disorder called leukocyte adhesion deficiency type 1 (LAD-1). LAD-1 is characterized by impaired leukocyte adhesion and migration, resulting in recurrent bacterial infections and delayed wound healing. Integrin β2 has also been implicated in various inflammatory and immune-mediated diseases, including inflammatory bowel disease, rheumatoid arthritis, and asthma. Biochem/physiol Actions
Clone 1D15 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Integrin β2 (CD18). It targets an epitope within 17 amino acids from the N-terminal, extracellular domain.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
