General description
Transcription factor AP-2 gamma (UniProt: Q92754; also known as AP2-gamma, Activating enhancer-binding protein 2 gamma, Transcription factor ERF-1) is encoded by the TFAP2C gene (Gene ID: 7022) in human. Transcription factor AP-2 gamma (AP-2 ) is a critical sequence-specific DNA-binding protein, belonging to the AP-2 family, involved in regulating gene expression during embryonic development and promoting cell proliferation and survival in cancerous tissues. AP-2 comprised of an N-terminal transactivation domain and a C-terminal DNA-binding and dimerization domain, enabling it to interact with enhancer elements and form homo- or heterodimers with other AP-2 family members. It binds to the consensus sequence 5′-GCCNNNGGC-3′, influencing the transcription of genes pivotal for development and differentiation. It is a retinoic acid-responsive gene involved in placental development and the progression of human breast cancer. AP-2 is required for development and maintenance of extra-embryonic membranes, trophectoderm. AP-2 is predominantly localized in the nucleus, where it interacts with various proteins, including proto-oncogene PELP1, enhancing signaling pathways such as RET, which are crucial in tumorigenesis. Its expression is notably high in tissues such as the placenta and mammary glands, and aberrant expression has been linked to breast cancer progression and therapy resistance. AP-2 is involved in pathways such as the regulation of the estrogen receptor and HER-2/neu proto-oncogene, implicating it in diseases like breast cancer and Char syndrome. Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected AP-2 in T-47D cell lysate.
Affinity Binding Assay: A representative lot of this antibody bound with AP-2 peptide with at least one thousand-fold (1,000X) higher affinity than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected AP-2 in human skin tissue sections.

Biochem/physiol Actions
Clone 2J5 is a ZooMAb® rabbit recombinant monoclonal antibody that detects AP-2 . It targets an epitope within 26 amino acids from the C-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
0.3 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
