General description
ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. E3 UFM1-protein ligase 1 (UniProt: O94874; also known as E3 UFM1-protein transferase 1, EC:2.3.2.-, Multiple alpha-helix protein located at ER, Novel LZAP-binding protein, Regulator of C53/LZAP and DDRGK1) is encoded by the UFL1 (also known as KIAA0776, MAXER, NLBP, RCAD) gene (Gene ID: 23376) in human. UFL1 plays a crucial role in the ubiquitin-proteasome system, mediates ufmylation and function as a ubiquitin-like modifier. It is involved in regulating diverse biological processes including ER homeostasis, ribosome recycling (e.g., ufmylating ribosomal proteins such as RPL26), DNA damage response, autophagy, and immune signaling. UFL1 is a ~794 amino acid (~90 kDa) scaffold‑type E3 ligase and, its N‑terminal helix is highly conserved and essential for binding to the E2 enzyme UFC1, facilitating the transfer of UFM1 to substrates. Expression profiling indicates ubiquitous presence across tissues, with high expression in liver. UFL1 is primarily localized in the endoplasmic reticulum (ER) membrane via interaction with DDRGK1/UFBP1. Additionally, localizes in the cytoplasm, nucleus and at chromatin, particularly at DNA double‑strand break sites and telomeres suggesting multifaceted roles in genome maintenance. UFL1 is involved in ER stress responses, hematopoiesis, pancreatic and plasma cell function, and myocardial homeostasis. Deficiency of UFL1 provokes ER stress, unfolded protein response (UPR), and apoptosis in secretory and hematopoietic cells, impairs cardiomyocyte function leading to dilated cardiomyopathy, and disrupts DNA damage response by affecting MRE11 and histone H4 ufmylation, compromising ATM activation. UFL1 also modulates antiviral immunity by maintaining STING stability (via TRIM29 inhibition) in a ufmylation‑independent manner, enhancing interferon‑β responses during DNA virus infection. Biochem/physiol Actions
Clone 2B9 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects UFL1. It targets an epitope within 18 amino acids from the C-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
