General description

POlyhedrin delivery systems (PODS) are a class of delivery systems specifically designed for the efficient delivery of proteins to target cells or tissues. They offer a platform to overcome the challenges associated with protein delivery, such as stability, cell membrane permeability, and targeted delivery. We offer Protein delivery system, empty, PODS which are empty crystals composed solely of polyhedrin protein which self-assembles into regular, cubic crystals. They do not contain any cargo protein, and are intended to be used as negative controls alongside cargo-containing PODS co-crystals. The cross-reactivity of the product has been tested on a variety of cells including mouse ES cells, PC12, fibroblasts, and ETS embryos, and there was no observable interference with the tested cells. Additionally, in-vivo data indicate no inflammatory response to PODS empty crystals in animals.

Application

Protein delivery system, empty, PODS is a POlyhedrin Delivery System (PODS). It can be used in research areas such as biocatalysis, drug delivery systems, gene therapy, targeted therapy, tissue engineering and regenerative medicines. The product is intended to be used as negative controls alongside cargo-containing PODS co-crystals which contain cargo proteins. PODS are also highly stable in storage which provides a unparalleled flexibility and control of a culture system and opens up many possibilities for innovation in 2D, 3D and in-vivo applications. The product can also be used for studies in gene therapy approaches where it requires to deliver gene-editing proteins, such as CRISPR-Cas9, to specific cells or tissues to modify the genetic material and treat genetic disorders or other diseases. In drug delivery applications, the product can be used to deliver various therapeutic proteins, such as cytokines, growth factors, antibodies, and enzymes.

Features and Benefits

  1. This product is produced with no animal derived raw products. All processing and handling employs animal free equipment and animal free protocols.
  2. The active protein is captured in its nascent, natively folded form with limited scope for proteolytic degradation. Consequently, excellent levels of bioactivity are observed.
  3. Formulation: lyophilized from a volatile solution.
  4. The product comes with a molecular weight of 28.7 kDa, endotoxin level of <0.06 EU/ml, are stable for at least 1 year when dry and 6 months when resuspended.
  5. PODS® co-crystals may be reconstituted at 200 million co-crystals/ml in sterile PBS. 20% glucose has a buoyant density closer to PODS co-crystals and can be useful for aliquoting. PODS co-crystals are highly stable when stored in aqueous solution (pH range 6 - 8).
  6. PODS provides durability, stability AND sustained release, addressing the limitations of conventional and enabling researchers to effortlessly control and manipulate cells in 2D and 3D culture as well as in-vivo.
