General description
Apolipoproteins B (apoB) is a large (4538 amino acids), amphipathic glycoprotein. It is encoded by the gene APOB, mapped to human chromosome 2. A unique mRNA editing process results in the formation of two structurally related by distinct isoforms of apo-B, includes apoB-48 and apoB-100. ApoB-100 is synthesized in the liver. apoB has both hydrophobic lipid binding regions and hydrophilic region that interacts with plasma. It is arranged as α-helices and β-sheet domains, designated NH2-βα1-β1-α2-β2-α3-COOH. The mature protein has 25 cysteine residues, out of which 16 residues are involved in intramolecular disulfide bridges and proper folding of the amino terminal end of the protein. MB47 binds near to 3500 amino acid of apoB-100.
Biochem/physiol Actions
Apolipoproteins B (apoB) plays a key role in mammalian lipoprotein metabolism. ApoB-100 is the structural protein involved in triglyceride rich very low sdensity lipoprotein (VLDL) synthesis. apo-B100 is the protein in cholesteryl ester-enriched low-density lipoprotein (LDL). High plasma levels of apo-B100 is involved in premature atherosclerosis. apo-B100 functions as a ligand for receptor mediated endocytosis of LDL.
