General description

The 3-sn-phosphatidylinositol-4-phosphate (PIP) and lipid A adjuvant, are component of WR304. WR304 mAb-biotinylated antibody was developed by the Walter Reed Army Institute of Research (WRAIR).
WR304 is an antibody that binds specifically to the lipids PIP and PIP2. WR304 also neutralizes infectious HIV-1 virus and can be used to probe the presence of PIP and PIP2 in membranes in a variety of applications including inositol phosphate-dependent cell signaling pathways. WR304-Biotin allows this antibody to be used in combination with a wide range of commercially available streptavidin-fluorophore and streptavidin-enzyme complexes. WR304-Biotin can also be used with samples that contain mouse IgM.This antibody was developed by the Walter Reed Army Institute of Research (WRAIR) using a targeted adjuvant protocol with liposomes containing Avanti′s Lipid A.

Biochem/physiol Actions

WR304 is an antibody that binds specifically to the lipids 3-sn-phosphatidylinositol-4-phosphate (PIP) and PIP2. WR304 neutralizes infectious human immunodeficiency virus (HIV-1 virus). WR304 interacts with sulfatide and not with neutral glycolipids. It is effective in neutralizing human peripheral blood mononuclear cells (PBMC) associated infections. WR304 can be used to probe the presence of PIP and PIP2 in membranes in a variety of applications including inositol phosphate-dependent cell signaling pathways. WR304-Biotin allows this antibody to be used in combination with a wide range of commercially available streptavidin-fluorophore and streptavidin-enzyme complexes. WR304-Biotin can also be used with samples that contain mouse IgM.

Two human mAbs (2F5 and 4E10), originally derived from HIV-1-infected patients, are important, but rare, mAbs that exhibit broad cross-clade neutralizing activities against HIV-1. In addition to peptide sequences on the gp41 envelope protein, both antibodies reportedly also bound specifically

The binding characteristics of two monoclonal antibodies (mAb) to phosphatidylinositol-4-phosphate (PIP) were examined: a murine IgM mAb to PIP; and a human IgG mAb (4E10) that binds both to HIV-1 envelope protein and also to neutral and anionic phospholipids, including

Monoclonal antibodies against phosphatidylinositol phosphate were produced after injecting a mouse with liposomes containing dimyristoylphosphatidylcholine, cholesterol, phosphatidylinositol phosphate and lipid A. The antibodies raised were IgM (kappa) and their activities were assayed by complement-dependent damage to liposomes lacking lipid A
