General description
Monophosphoryl lipid A (MPLA) is produced by hydrolysis of endotoxin lipopolysaccharide (LPS) isolated from Salmonella minnesota R595. MPLA is purified from the hydrolysate and contains a mixture of structural analogs containing differing numbers of fatty acids and positional isomers.
Purified Lipid A (MPLA) is soluble in DMSO at ~1mg/mL. Alternatively, it has been reported that MPLA can be suspended at ~1mg/mL in water or isotonic saline containing 0.2% triethylamine. After adding water/saline containing triethylamine to the MPLA powder, swirl the suspension in 65-70°C water bath for 10-20 seconds, then disperse by sonication using an ultrasonic bath for 30 seconds. The final solution will be slightly turbid.
Application
Lipid A - purified has been used as an adjuvant in the reconstitution lipid mixture.
Lipid A- purified has been used for toll-like receptor (TLR) ligand treatment.
Biochem/physiol Actions
Detoxified monophosphoryl lipid A (MPLA) acts as a toll-like receptor 4 (TLR4) agonist. Hydrolysis of the lipopolysaccharide (LPS) to yield MPLA, retains the action of stimulating the adaptive immune response while eliminating many of the pro-inflammatory side effects experienced with LPS.
