Physical form

Anti-hnRNPA1 (Mouse Monoclonal), . One vial containing 50 μg of purified antibody in 75 μL of 0.1 M Tris-Glycine (pH7.4), 150 mM NaCl with 0.05% sodium azide and 30% glycerol. Store at -20°C .
Normal Mouse IgG,. One vial containing 125 µg purified mouse IgG in 125 µL storage buffer containing 0.1% sodium azide. Store at -20°C.
RIP Primers, E2F3 . One vial containing 75 μL of 5 μM of each primer specific for human E2F3 mRNA.
Store at -20°C.

Preparation Note

Stable for 1 year at -20°C from date of receipt.Handling Recommendations: Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.

Analysis Note

RNA Binding Protein Immunoprecipitation:
RIP Lysate prepared from HeLa cells (2 X 10E7 cell equivalents per IP) were subjected to immunoprecipitation using 5 µg of either a normal mouse IgG ), or 5 µg Anti-hnRNPA1 antibody (Part. # CS204408) and the Magna RIP RNA-Binding Protein Immunoprecipitation Kit (Cat. # 17-700).
Successful immunoprecipitation of hnRNPA1-associated RNA was verified by qPCR using RIP Primers E2F3, (Figure 1).

FUNCTION: Involved in the packaging of pre-mRNA into hnRNP particles, transport of poly(A) mRNA from the nucleus to the cytoplasm and may modulate splice site selection. May play a role in HCV RNA replication.

Interacts with SEPT6. Interacts with HCV NS5B and with the 5′-UTR and 3′-UTR of HCV RNA.

SUBCELLULAR LOCATION: Nucleus. Cytoplasm. Note: Localized in cytoplasmic mRNP granules containing untranslated mRNAs. Shuttles continuously between the nucleus and the cytoplasm along with mRNA. Component of ribonucleosomes. In the course of viral infection, colocalizes with HCV NS5B at speckles in the cytoplasm in a HCV-replication dependent manner.

PTM: Arg-194, Arg-206 and Arg-225 are dimethylated, probably to asymmetric dimethylarginine.

SEQUENCE SIMILARITIES: Contains 2 RRM (RNA recognition motif) domains.
Includes negative control mouse IgG antibody and control primers specific for the mRNA of human E2F3.
