General description
The synergistic effect of the indicator dyes, Eosin Y and Methylene Blue differentiates colonies of lactose fermenting and non-fermenting microbes. Growing colonies of E. coli produce a green metallic sheen due to high acid production from the medium.
Other coliforms do not produce enough acid to attain the metallic sheen. The growing coliforms produce blue-black colonies due to the formation of eosin-methylene blue dye complex when the pH drops.
Peptones in the medium provide nitrogen, amino acids, and minerals to support growth. Di-potassium phosphate acts as a buffer and agar is the solidifying agent.
