General description

Adeno-associated virus (AAV) is recognized as a preferred vector for gene therapy due to its infectivity, versatility, and favorable safety profile. The various AAV serotypes exhibit distinct tissue specificities, transduction efficiencies, and immunogenicity, allowing for tailored approaches to gene therapy. While AAV generally has low immunogenicity, exposure to these viruses is common which leads to a high rate of seroprevalence in the general population. Pre-existing antibodies against AAV can challenge a gene therapy’s safety and efficacy making it essential to assess seroprevalence and immunogenicity in descriptive preclinical testing.

Application

MILLIPLEX® Qualified assays undergo rigourous assay development, verification, and Quality Control testing to achieve optimal performance. Simultaneously analyze up to 6 anti-AAV antibodies in human serum and plasma.

Analytes included: anti-AAV1, anti-AAV2, anti-AAV5, anti-AAV6, anti-AAV8, anti-AAV9

Assay Characteristics: Refer to kit protocol for assay cross-reactivity and sensitivity.

Features and Benefits

  • Pioneering Technology: This first of its kind assay provides researchers provides a powerful tool to study immune responses to various AAV serotypes.
  • Multiplex Detection: Simultaneously detects antibodies to multiple AAV serotypes (AAV1, AAV2, AAV5, AAV6, AAV8, & AAV9) plus 4 assay control beads.
  • Complete Solution: Includes all necessary reagents and an intuitive protocol thus streamlining the workflow and reducing the need for multiple assays.
  • Demonstrated Precision and Specificity: Ensures high reliability with <10% intra-assay CV and <15% inter-assay CV, while specificity testing with serotype-specific control antibodies confirmed negligible cross-reactivity to non-specific antigens in most cases.

Preparation Note

Recommended storage for kit components is 2-8˚C.
