Western blot: 1-1.5 μg/mL (1:200:1:300) using ECL

Dilutions should be made using a carrier protein such as BSA (1-3%). Typical bands detected are ~125kDa in rat membrane preparations. Excellent methods given in Ling, et al (2003) {http://jp.physoc.org/cgi/content/full/553/1/65}

Optimal working dilutions must be determined by the end user.

Biochem/physiol Actions

Corresponds to the C-terminus of the longer form of BKCa channel a subunit and overlaps by >40 residues with a shorter form.

Physical form

Affinity purified immunoglobulin. Lyophilized from phosphate buffered saline, pH 7.4, containing 1% BSA, 5% sucrose as a stabilizer and 0.025% sodium azide as a preservative. Reconstitute with 50 μL of sterile deionized water. Centrifuge antibody preparation before use (10,000 xg for 5 min).

Preparation Note

Maintain lyophilized material at -20°C for up to 12 months. After reconstitution maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

Analysis Note

Control
CONTROL ANTIGEN: Included free of charge with the antibody is 120 μg of control antigen (lyophilized powder in phosphate buffered saline, pH 7.4, containing 5% sucrose and 0.025% sodium azide). The stock solution of the antigen can be made up using 100 μL of sterile deionized water. For positive control, in Western blotting, use 20 ng protein per lane. For negative control, preincubate 3-5 μg of purified peptide with 1 μg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.

Fibroblast-like synoviocytes (FLSs) are a key cell type involved in rheumatoid arthritis (RA) progression. We previously identified the KCa1.1 potassium channel (Maxi-K, BK, Slo 1, KCNMA1) as a regulator of FLSs and found that KCa1.1 inhibition reduces disease severity in

Large-conductance Ca(2+)-activated K(+) (BK(Ca)) channels encoded by the Slo1 gene are ubiquitously expressed, and they play a role in regulation of many cell types. In excitable cells, BK(Ca) channels and voltage-activated Ca(2+) channels often form functional complexes that allow the
