General description
Histone H3.1 UniProt: Q16695; also known as H3/t, H3t, H3/g) is encoded by H3-4 (also known as H3FT, HIST3H3) gene (Gene ID: 8290) in human. Histone H3 has two main variants, H3.1 and H3.3, which show different genomic localization patterns in animals. Histone H 3.1 is a core component of nucleosome that is present only in mammals. It is expressed during S phase, then its expression decreases significantly as cell division slows down during the process of differentiation. Histone H 3.1 expression is shown to be replication dependent. It serves as the canonical histone, which is incorporated during DNA replication. Histone 3.1 can also undergo serotonylation of glutamine at position 5 (Q5ser) in organisms that produce serotonin. This has been demonstrated that tissue transglutaminase 2 can serotonylate histone H3K4me3 marked nucleosomes, which results in the formation of H3K4me3Q5ser. The presence of Q5ser does not affect the ability of MLL1, an H3K4 methyltransferase, to modify nucleosomes in vitro. In the brain the H3K4me3Q5ser signal is not segregated only to regions in which serotonin is produced, but is shown to be broadly distributed. H3K4me3Q5ser expression has also been reported in human pluripotent stem cell-derived serotonergic neurons and with differentiation the levels of H3K4me3Q5ser increase significantly. Western Blotting Analysis: A 1:500 dilution from a representative lot detected Histone H3 trimethylated at Lys4 and serotonylated at Gln5 in whole cell lysate from differentiated RN46A-B14 cells but not in lysate from undifferentiated RN46A-B14 cells.
ChIP-Seq Analysis: A representative lot of this antibody was used to assess genome-wide impacts of serotonergic differentiation in differentiated RN46A-B14 cells (Farrelly, LA., et al. (2109). Nature. 567(7749); 535-539).
