General description

PIWI-L1, also known as HIWI, is a member of the PIWI subfamily of Argonaute proteins. PIWI-L1 has both PAZ and Piwi motifs, which are critical for the biogenesis and function of small non-coding RNAs. PIWI-L1 plays a central role in spermatogenesis and maintaining germline integrity by repressing transposable elements and preventing their mobilization. Additionally, it has been shown that PIWI-L1 plays an important role in RNA silencing, stem cell self-renewal, and translational regulation in diverse organisms. Recent studies have shown that overexpression of PIWI-L1 is a potential biomarker for colon, gastric and other cancers. Other studies have shown that removing PIWI-L1 protein from single germline stem cells significantly decreases the rate of their division, suggesting that PIWI-L1 regulates the self-renewal capacity of the germline and hematopoietic stem cells. PIWI-L1 is detected in most fetal and adult tissues, as well as spermatocytes and spermatids during spermatogenesis. Further, it is expressed highly in testicular tumors originating from embryonic germ cells, but not in testicular tumors of somatic origin.
~120 kDa observed. Uncharacterized bands may appear in some lysates. Uniprot describes 3 isoforms between ~90 kDa and ~99 kDa
