Brain and acute leukemia cytoplasmic protein (UniProt: Q8WXS3; also known as BAALC) is encoded by the BAALC gene (Gene ID: 79870) in human. BAALC is predominantly expressed in neuroectoderm-derived tissues. Its expression has been reported in the brain and spinal cord, and at low levels, in the adrenal gland. In the bone marrow, it is shown to be confined to the CD34+ progenitor cells. In neurons, it localizes to postsynaptic lipid rafts and in myocardial and skeletal muscle cells, it is shown to be localized to the cytoplasm adjacent to the inner cell membrane. Overexpression of BAALC has been reported in subjects with acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL). Its high expression is considered as an independent marker of poor prognosis. In leukemia cells, BAALC induces cell-cycle progression by sustaining ERK activity through an interaction with a scaffold protein MEK kinase-1 that blocks the interaction between ERK and MAP kinase phosphatase 3. It is shown to confer chemoresistance in AML cells by upregulating ATP-binding cassette proteins in an ERK-dependent manner and blocks ERK-mediated monocytic differentiation of AML cells by trapping KLF4 in the cytoplasm and inhibiting its function in the nucleus.
