General description

Major intrinsically disordered NOTCH2-binding receptor 1-like (UniProt: P59773; also known as Membrane integral NOTCH2-associated receptor 2) is encoded by the MINAR2 (also known as KIAA1024L) gene (Gene ID: 100127206) in human. MINAR2, a highly conserved, intrinsically disordered member of the MINAR family of proteins, has an extracellular domain, a single transmembrane domain, and a few amino acids in its cytoplasmic tail. Its expression is observed in the brain with high levels detected in midbrain, hypothalamus, and the pituitary gland. Loss of MINAR2 is reported to cause motor dysfunctions in murine models that resemble Parkinson′s disease. MINAR2 knockout mice display significant alterations in their neuronal function, loss of TH positive neurons in pars compacta, and higher accumulation of -synuclein protein. It is reported to be localized to endoplasmic reticulum (ER) compartments and interacts with NOTCH2. ER stress, NOTCH folding, and its trafficking from the ER have been proposed to play a role in motor deficits associated with progression of Parkinson s disease. MINAR2 has also been linked to progressive hearing loss with alteration sin the stereocilia of hair cells. Loss of MINAR2 is shown to reduce cholesterol localization to the hair bundles in the ear that aggravates hair cell deficits. (Ref.: Bademci, G., et al. (2022) Proc. Natl. Acad. Sci. Immunogen

A linear peptide corresponding to 14 amino acids from the C-terminal half of human Membrane integral NOTCH2-associated receptor 2 (MINAR2).

Application

Quality Control Testing

Evaluated by Western Blotting in lysates from HEK293 cells transfected mouse MINAR2.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected mouse MINAR2 in lysates from HEK293 cells transfected mouse MINAR2, but not in lysates from untransfected cells.

Tested Applications

Immunohistochemistry Applications: A representative lot detected MINAR2/KIAA1024L in Immunohistochemistry application (Ho, R., et. al. (2020). Brain Commun. 2(1):fcaa047).

Immunocytochemistry Analysis: A representative lot detected MINAR2/KIAA1024L in Immunocytochemistry applications (Ho, R., et. al. (2020). Brain Commun. 2(1):fcaa047).

Preparation Note

1.0 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
