General description

AMPylation is a reversible post-translational modification in which adenosine monophosphate (AMP) derived from ATP is added to threonine and tyrosine (and possibly serine) residues on target proteins. This modification is created by proteins containing Fic (filamentation induced by cyclic adenosine monophosphate) domains, which are present in both prokaryotic and eukaryotic cells. In mammals, the HYPE protein is the most widely known Fic-containing protein and the precise function of its AMPylated targets is not yet clear. However, bacterial cells possess numerous Fic-domain proteins that modify and disable protein targets in host cells, thereby suppressing signaling pathways and increasing the robustness of pathogenic infections. The Vibrio parahaemolyticus VopS protein, for instance, disrupts the function of Rho guanine triphosphatases, and eventually disrupts the actin cytoskeleton.

Preparation Note

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
