General description

Histone-lysine N-methyltransferase SETDB1 (UniProt: Q15047; also known as EC:2.1.1.366, ERG-associated protein with SET domain (ESET), Histone H3-K9 methyltransferase 4 (H3-K9-HMTase 4), Lysine N-methyltransferase 1E, SET domain bifurcated 1) is encoded by the SETDB1 (also known as ESET, KIAA0067, KMT1E) gene (Gene ID: 9869) in human. SETDB1, is a histone methyltransferase, member of the Suppressor of Variegation 3 9 (SUV39)-related protein lysine methyltransferases, that plays a key role in the epigenetic regulation of gene expression, chromatin remodeling, and implicated in the regulation of cell cycle progression, inactivation of the X chromosome, immune cells function and nervous system development. Structurally, SETDB1 contains several functional domains, including three N-terminal tandem Tudor domain, that recognizes histone H3 sequences containing both methylated and acetylated lysines, and a methyl-CpG-binding domain (MBD). The C terminal region harbors a pre-SET, a bifurcated SET, and a post-SET domain that are required for its methyltransferase activity. The N-terminal region also contains two nuclear export signals (NES) and two nuclear localization signals (NLS), that regulate SETDB1 localization. SETDB1 specifically tri-methylates lysine 9 on histone H3 (H3K9me3), a mark associated with transcriptional repression. This modification is crucial for maintaining heterochromatin structure and silencing endogenous retroviruses and other repetitive elements. SETDB1 is ubiquitously expressed, with particularly high levels in embryonic stem cells and certain adult tissues, reflecting its role in maintaining pluripotency, regulating developmental processes and silencing euchromatic genes. Involvement in disease is significant, as aberrant expression or mutations of SETDB1 have been implicated in the pathogenesis of gliomas and various cancers, including melanoma, lung and breast cancer, gastrointestinal and ovarian tumors. It can contribute to oncogenesis by silencing tumor suppressor genes. Additionally, SETDB1 dysregulation is linked to neurological disorders, such as Huntington′s disease and schizophrenia. Recombinant baculovirus gp64-fusion protein containing amino acids 56 amino acids from the C-terminal half of human SETDB1.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.
