General description

CRISPR-associated endodeoxyribonuclease Cas12f1 (UniProt: A0A482D308; EC:3.1.-.-; also known as Un1Cas12f1, CRISPR-associated endodeoxyribonuclease Cas14a1) is encoded by the cas12f (also known as cas14a1) gene in Archaea. CRISPR-Cas systems are adaptive immune mechanisms found in bacteria and archaea that provide protection against mobile genetic elements (viruses, transposable elements and conjugative plasmids). The acronym CRISPR stands for "Clustered Regularly Interspaced Short Palindromic Repeats," which are DNA sequences that store segments of viral DNA, allowing the organism to recognize and defend against future infections. Cas12f1 protein consists of 529 residues. The amino-terminal domain (NTD) and contains a wedge (WED), a recognition (REC), and a zinc finger (ZF) domain. The carboxy-terminal domain (CTD) consists of the RuvC domain and another ZF domain, and a termed the target nucleic acid-binding (TNB) domain. The miniature Cas system (CasMINI) was engineered from the type V-F Cas12f (Cas14) system, and its activity is comparable to Cas12a for gene activation. It is highly specific and suitable for gene editing in mammalian cells with the promise of effective gene therapy applications. (Ref.: Xu, X., et al., (2021). Mol. Application

Quality Control Testing

Evaluated by Western Blotting in HEK293T cells stably transfected with 3xFLAG-tagged CasMini.

Western Blotting Analysis: A 1:500 dilution from a representative lot detected CasMini in HEK293T cells stably transfected with 3xFLAG-tagged CasMini.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 8H3-H1 is a mouse monoclonal antibody that detects CasMini, N-terminal fragment. It targets an epitope within 135 amino acids from the N-terminal region.

Physical form

Purified mouse monoclonal antibody IgG1 in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.

0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
