General description

Zinc finger protein ZFAT (UniProt: Q7TS63; also known as Zinc finger protein 406) is encoded by the Zfat (also known as Gm922, Zfat1, Zfp406, Znf406) gene (Gene ID: 380993) in mouse. ZFAT is a transcription factor and is suggested to be involved in lymphocyte signaling. Human homolog ZFAT (also known as zinc finger gene in AITD susceptibility region) is strongly expressed in placenta, spleen, kidney, testis and peripheral blood leukocytes. It is also expressed in CD4+ and CD8+ T-cells, CD19+ B-cells and CD14+ monocytes. ZFAT is primarily localized in the nucleus, where it functions as a transcription factor, essential for regulating gene expression and is also found in the cytosol. Zinc finger protein ZFAT contains multiple zinc finger motifs that facilitate its interaction with DNA, enabling specific binding to target gene sequences. Four isoforms of ZFAT have been described that are produced by alternative splicing. In immune related cells, ZFAT is proposed to control apoptosis and cell survival. It also participates in endothelium organization and capillary formation. Studies demonstrated that Zfat regulates the transcription of target genes by binding directly to the TSS proximal region, and that Zfat-target genes play important roles in T-cell homeostasis. Overexpression of ZFAT causes down-regulation of a number of genes involved in the immune response. ZFAT is down-regulated in preeclampsia. Polymorphism in ZFAT is linked with severity in Hashimoto s disease (HD). Quality Control Testing

Evaluated by Western Blotting in HEK-293 cells transfected with mouse ZFAT.

Western Blotting Analysis: A 1:100,000 dilution from a representative lot detected ZFAT in HEK-293 cells transfected with mouse ZFAT but not in HEK-293 cells transfected with human ZFAT.

Biochem/physiol Actions

Clone Z4 is a rat monoclonal antibody that specifically detects ZFAT. It targets an epitope within 164 amino acids from the C-terminal region.

Purified rat monoclonal antibody IgG2a in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.
