General description

DNA helicase MCM8 (Uniprot; Q9UJA3; also known as Minichromosome maintenance 8) is encoded by MCM8 gene (Gene ID; 84515) in human. It is a nuclear protein, localizes to nuclear foci and to double-stranded DNA breaks. Its expression is highest in placenta, lung and pancreas and low in skeletal muscle and kidney. Also, expressed in various tumors with highest levels in colon and lung cancers. MCM8 is a member of MCM proteins, which are key components of the pre-replication complex, recruit replication-related proteins and form replication forks. It is a component of the MCM8-MCM9 complex which is involved in the repair of double-stranded DNA breaks (DBSs) and DNA interstrand cross-links (ICLs) by homologous recombination (HR). MCM8 is particularly important in the restart of stalled replication forks, contributing to the maintenance of genome stability. Additionally, it has been associated with meiotic recombination and DNA repair processes, highlighting its significance in preserving the integrity of the genetic material. The dysregulation of MCM8 has been linked to genomic instability and cancer development, emphasizing its role as a potential target for cancer therapy and as a biomarker for assessing genomic instability. KLH-conjugated linear peptide corresponding to 18 amino acids from the N-terminal half of human MCM8.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell nuclear extract.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MCM8 in HeLa cell nuclear extract.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MCM8 in HEK293T and A-431 cell lysates.

Clone 2D4 is a mouse monoclonal antibody that specifically detects MCM8 protein. It targets an epitope within 18 amino acids from the N-terminal half of the protein.

Purified mouse monoclonal antibody IgG2a in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.
