General description

Killer cell lectin-like receptor 8 (UniProt: Q60682; also known as Lymphocyte antigen 49h, Ly-49h, T-cell surface glycoprotein Ly-49H) is encoded by the Klra8 (Gene ID: 16639) in murine species. Ly-49H is a disulfide-linked, homodimeric, single-pass type II membrane glycoprotein that contains a cytoplasmic domain (aa 1-44), a transmembrane domain (aa 45-66), and an extracellular domain (aa 67-266). Its C-type lectin domain is localized in amino acids 143-261. Ly-49H is expressed on a subset of natural killer (NK) cells and serves as a receptor for class I HMC. It can also be co-expressed with Ly-49 inhibitory receptors. However, unlike Ly-49 inhibitory receptors, Ly-49H is not detectable on naive splenic CD31 T cells, indicating that Ly-49H is an NK cell-specific activation receptor. It lacks the immunoreceptor tyrosine-based inhibitory motifs (ITIM) in its cytoplasmic domain but associates with DAP12 that possesses an immunoreceptor tyrosine-based activation motif (ITAM). Its interaction with DAP2 leads to NK cell activation. Clone 3D10 specifically binds Ly-49H and does not cross-react with Ly-49A, C, D, and G2. It abolishes Ly-49H-dependent activation of NK cells. Application

Quality Control Testing

Evaluated by Flow Cytometry in C57BL/6 mouse splenocytes.

Flow Cytometry Analysis: 1 µg of this antibody detected Ly49H/KLRA8 in one million C57BL/6 mouse splenocytes.

Tested Applications

Immunoprecipitation Analysis: A representative lot immunoprecipitated Ly-49H/KLRA8 in Immunoprecipitation applications (Smith, H.R., et al. (2000). J Exp Med. 191(8):1341-54).

Inhibition Assay: A representative lot of this antibody abolished Ly49H-dependent activation of NK cells.(Forbes, C.A., et al. (2014). PLoS Pathog. 10(5):e1004161).

Flow Cytometry Analysis: A representative lot detected Ly-49H/KLRA8 in Flow Cytometry applications (Smith, H.R., et al. (2000). J Exp Med. 191(8):1341-54; Forbes, C.A., et al. (2014). PLoS Pathog. 10(5):e1004161).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 3D10 is a mouse monoclonal antibody that detects Killer cell lectin-like receptor 8 (KLRA8/Ly-49H). It targets an epitope within the extracellular domain.

Preparation Note

1.0 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
