General description

CD151 antigen (UniProt: P48509; also known as GP27, Membrane glycoprotein SFA-1, Platelet-endothelial tetraspan antigen 3 (PETA-3), Tetraspanin-24 (Tspan-24)) is encoded by the CD151 (also known as TSPAN24) gene (Gene ID: 977) in human. CD151 is a transmembrane glycoprotein belonging to the tetraspanin family, characterized by four hydrophobic transmembrane domains and two extracellular loops that mediate protein-protein interactions. CD151 acts as a scaffold protein, facilitating the organization of integrin receptors, particularly laminin-binding integrins such as 3β1, 6β1, and 6β4, into specialized tetraspanin-enriched microdomains on the plasma membrane. CD151 plays a pivotal role in cell adhesion, migration, and signaling by modulating integrin-mediated interactions with the extracellular matrix. It is highly expressed in epithelial cells and endothelial cells, with elevated levels observed in various malignancies. CD151 is predominantly localized at intercellular junctions in normal tissues, whereas its cytosolic localization in cancer cells correlates with metastatic potential. CD151 interacts with growth factor receptors such as EGFR and HGFR, influencing pathways like PI3K/Akt and NF- B, which are critical for tumor progression and metastasis. Dysregulation of CD151 contributes to enhanced tumor invasion, angiogenesis, and metastasis in cancers such as lung, prostate, and hepatocellular carcinoma.
