General description

CD226 antigen (UniProt: Q15762; also known as DNAX accessory molecule 1 (DNAM-1)) is encoded by the CD226 (also known as DNAM1) gene (Gene ID: 10666) in human. DNAM-1/CD226, is an immunoglobulin superfamily member that functions as a co-stimulatory receptor on various immune cells, including natural killer (NK) cells, T cells, and some myeloid cells. DNAM-1/CD226 is characterized by its extracellular Ig-like C2-type domains (aa 19-126 and aa 135-239), which facilitates its interaction with ligands such as CD112 and CD155. This interaction is crucial for mediating NK cell and cytotoxic T lymphocyte (CTL) activities against tumor cells and virus-infected cells. DNAM-1/CD226 is predominantly expressed on NK cells, CD8+ T cells, and subsets of CD4+ T cells. DNAM-1/CD226 plays a role as a co-stimulatory molecule is pivotal in immune surveillance, particularly in the context of tumor immunology. DNAM-1′s interaction with its ligands facilitates the activation and proliferation of NK and T cells, thus promoting the clearance of malignant cells. DNAM-1/CD226 has been implicated in various cancers and autoimmune disorders. Its expression and function are often altered in malignancies, affecting the immune system′s ability to recognize and destroy tumor cells. Loss of DNAM-1/CD226 contributes to CD8+ T cell exhaustion in chronic HIV-1 infection.
