General description
Tau-tubulin kinase 1 (EC 2.7.11.1; UniProt Q5TCY1; also known as Brain-derived tau kinase) is encoded by the TTBK1 (also known as BDTK, KIAA1855) gene (Gene ID 84630) in human. Tau-tubulin kinase 1 and 2 (TTBK1 and TTBK2) belong to the casein kinase 1 superfamily of kinases. TTBK2 is ubiquitously expressed, while TTBK1 is neuron-specific and can directly phosphorylate tau proteins at multiple Ser/Thr residues that are found on paired helical filament-tau or PHF-tau in Alzheimer′s disease (AD) brains. Consistently, transgenic mice over-expressiong human TTBK1 develop age-dependent pathology in brain, including intraneuronal accumulation of phosphorylated neurofilament (p-NF), tau phosphorylation, cortical and hippocampal astro/microgliosis, calpain I up-regulation, enhanced CDK5/p35 complex formation, down regulation of NMDA receptor, and memory impairment.

Immunohistochemistry Analysis: A representative lot detected a much higher TTBK1 immunoreactivity in paraformaldehyde-fixed frozen hippocampus and cortex sections from mice with human TTBK1 transgene (Xu, J., et al. (2010). FASEB J. 24(8):2904-2915).
