Preparation Note

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Repulsive guidance molecule B (UniProt: Q7TQ33; also known as RG11-responsive axonal guidance and outgrowth of neurite, DRAGON) is encoded by the Rgmb gene (Gene ID: 68799) in mouse. It is a glycosylphosphatidylinositol-anchored protein, which plays a significant role in various biological processes that contributes to the patterning of the developing nervous system, neuronal ashesion and bone morphogenetic protein (BMP) signaling. RGMb is expressed in multiple tissues, with notable levels in the developing nervous system, adult dorsal root ganglion sensory neurons, spinal cord, and brain. RGMb protein is synthesized with a signal peptide (aa 1-48), propeptide (aa 416-436) which is subsequently cleaved off to produce the mature protein (aa 49-415). Subcellularly, RGMb is localized at the cell membrane, where it interacts with receptors such as neogenin, influencing cell migration and differentiation. Dysregulation of RGMb has been implicated in several diseases, including cancer, where it has been shown to promote tumor progression by enhancing macrophage infiltration and inflammation in tissues such as the kidneys. Studies have highlighted that RGMb′s interaction with TAB1 and TRAF6 can activate downstream signaling pathways, further contributing to its role in disease pathology. Research has demonstrated that programmed death ligand 2 (PD-L2 binds to both PD-1 (inhibits antitumor immunity), and to RGMb, which regulates respiratory immunity, targeting the PD-L2 pathway has therapeutic potential for asthma, cancer, and other immune-mediated disorders. It is suggested that this pathway may provide insights into how to optimally modulate the PD-1 pathway in cancer immunotherapy while minimizing adverse events. This 307.9D1 rat monoclonal antibody has been validated by Flow cytometry, ELISA, Western Blotting, Immunocytochemistry and can be used for inhibition studies.(Ref.: Xiao, Y., et al. (2014).
