Trefoil factor 3 (UniProt: Q07654; also known as TFF3, Intestinal trefoil factor, hITF, Polypeptide P1.B, hP1.B) is encoded by the TFF3 (also known as ITF, TFI) gene (Gene ID: 7033) in human. TFF3 is a small soluble peptide secreted by mucin-producing intestinal epithelial cells and goblet cells. It is also expressed in the uterus, breast, and in some parts of the hypothalamus and pituitary gland. TFF3 is synthesized with a signal peptide (aa 1-35), which is subsequently cleaved off to produce the mature form. It can exist as a monomer or disulfide-linked homodimer. Upon dimerization, the additional 3(10)-helix involving residues 53-55 occur in the TFF3 homodimer that contributes to its distinct structure. The monomeric form is shown to induce cell migration by activating the MAPK pathway independently of the epidermal growth factor and the homodimeric form is shown to have an anti-apoptotic effect on epithelial cells. TFF3 is reported to promote mucosal wound healing in mice and humans and its elevated expression has been linked with gastric adenocarcinoma progression and biliary diseases. TFF3 expression in the intestine is shown to be regulated by food intake and its overexpression is shown to improve glucose tolerance by blocking the expression of gluconeogenic enzymes.
