General description
3dGRO® Human iPSC-derived Liver Progenitor Organoids are derived from integration-free human induced pluripotent stem cells (iPSC, Cat No. SCC271) reprogrammed using the Simplicon RNA reprogramming technology (Cat. No. SCR550). Human iPSC cells were differentiated using a multi-step process from definitive endoderm to hepatic endoderm and eventually into liver progenitor organoids. These liver progenitor organoids are polarized self-organizing, 3D structures that can be expanded in long-term culture and cryo-preservable. These liver progenitor organoids show persistent growth and expansion post cryo-preservation. 3dGRO® human iPSC-derived liver progenitor organoids can be propagated long-term by passaging every 8-10 days using 3dGRO® Human Liver Progenitor Organoid Expansion Media (Cat. no. SCM313).
The 3dGRO® Human iPSC Derived Liver Organoid Progenitor express key liver progenitor markers HNF4a, SOX17, FOXA2 and SOX9, and showed proper apical and basal-lateral polarization marked by E-cadherin and ZO1 expression. 3dGRO® Human iPSC-derived Liver Progenitor Organoids can be differentiated into mature liver organoids containing functional hepatocytes and cholangiocytes when cultured in differentiation media for 14 days.

Preparation Note
Store in liquid nitrogen. The organoids can be cultured for at least 10 passages after initial thawing without significantly affecting the cell marker expression and functionality.
