General description

Reinforced Clostridial Agar (RCA) is a powdered microbiological culture medium designed for the cultivation and enumeration of Clostridia. It contains essential nutrients such as beef extract, casein enzymatic hydrolysate, yeast extract, and dextrose, which provide nitrogen, vitamins, amino acids, and carbon for microbial growth. Sodium acetate acts as a buffering agent, while L-cysteine hydrochloride serves as a reducing agent to support anaerobic conditions. The medium has a final pH of 6.8 ± 0.2 at 25°C and is non-selective for Clostridium spp. and Bacteroides spp.

Application

Reinforced Clostridial Agar is widely used in clinical testing, food and beverage microbiology for the cultivation and enumeration of Clostridia. It is suitable for anaerobic bacteria and is commonly employed in bioburden testing, agriculture, and personal care product microbiology.

Preparation Note

Suspend 51 g of Reinforced Clostridial Agar in 1000 ml of distilled water. Boil to dissolve the medium completely. Sterilize by autoclaving at 10 lbs. Pressure (115°C) for 15 minutes.
