General description

CelLytic Y Cell Lysis Reagent is versatile, phosphate-free and non-denaturing. It provides an effective method for cell lysis and protein solubilization. Target proteins maintain immunoreactivity or biological function. The protocol is brief and preformed at room temperature. No extreme conditions or glass beads are required. Use only 2.5-5 ml/gram yeast cells.

Application

CelLytic Y Cell Lysis Reagent has been used in lysis of yeast cells expressing DNA repair, Rad protein, heat shock protein (ssa1) and human interleukin 4 (hIL4).

  3. Can I use glass beads with CelLytic Y Cell Lysis Reagent, Product C4482?

    If cell growth conditions are not optimal, then glass beads can be used to increase yields.

  4. Should I use CelLytic Y Cell Lysis Reagent, Product C4482?

    For all cases in which the addition of DTT does not interfere with the application tested, it is recommended to add DTT (5 - 10 mM final concentration) to the CelLytic Y reagent.  Addition of DTT significantly improves total protein yield.

  5. Can I use CelLytic Y Cell Lysis Reagent, Product C4482, to make spheroplasts?

    No.  For spheroplast formation and protein extraction we recommend the CelLytic Y Plus kit.

  6. What is in CelLytic Y Cell Lysis Reagent, Product C4482?

    The formulation is considered proprietary, but we can state that it is a non-denaturing phosphate-free reagent.

  7. What strains have been used with CelLytic Y Cell Lysis Reagent, Product C4482?

    The efficiency of CelLytic Y reagent for protein extraction has been tested on different strains of Saccharomyces cerevisiae [Y187, Y190, W303(a), S288C, SP1, S1278b (diploid and haploid), BJ2168, and cdc25] and Schizosaccharomyces pombe.
