General description
Monoclonal Anti-Caspase 9 (mouse IgG1 isotype) is derived from the CAS9 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from a BALB/c mouse immunized with a synthetic peptide corresponding to amino acids of human caspase 9 protein, conjugated to keyhole limpet hemocyanin (KLH). Caspases (cysteine-requiring aspartate protease) are a family of proteases that share similarities in amino acid sequences, structure and substrate specificity. Caspases are normally present in the cell as inactive procaspases. Caspase 9 (also known as Mch6 and ICE-LAP6) gene encodes a protein product of 37 kDa (reported also as 46 kDa), which undergoes a cleavage upon activation, leading to p18/p15/p9 subunits. It is a member of the CED-3 subfamily, bearing high similarity to caspase. High level expression of caspase 9 is found in the heart, testis and ovary.

Biochem/physiol Actions
Caspase 9 is a downstream protease, activated in caspase 3- and granzyme B-mediated apoptosis. Procaspase 9 is activated when a complex is formed with the cofactor apoptotic protease activating factor-1 (Apaf-1) through the caspase recruitment domain (CARD). Caspase 9 plays a critical role in the survival of neuronal progenitors within the embryonic forebrain and cortex. Its deficiency leads to defective brain development and embryonic lethality. Overexpression of caspase 9, but not of a mutant in which the catalytic Cys was replaced with an Ala, induces apoptosis in MCF7 cells.
