Application

Cytochrome P450 Microsome Preparation Human is recombinant and prepared from Saccharomyces cervisiae expressing the human cytochrome 2C8 Isozyme coexpressed with human NADPH reductase and cytochrome b5. It is used to catalyze the oxidative metabolism of xenobiotics and to study the biotransformation and elimination of drugs and pollutants from the body. Cytochrome P450 is used to study hormone synthesis and breakdown, cholesterol synthesis, drug metabolism and bioactivation and vitamin D metabolism.

Biochem/physiol Actions

Microsomal cytochrome P450 enzymes are found primarily in the endoplasmic reticulum of liver tissue. They catalyze the oxidative metabolism of xenobiotics and are the major enzymes involved in drug metabolism and bioactivation. The reactions are achieved through a mixed monooxygenase system. The cytochrome P450 enzymes range in molecular mass between 45-60 kDa.

Physical form

Supplied as a solution containing 50 mM potassium phosphate, pH 7.4, 5% glycerol, 1 mM EDTA, 1 mM PMSF, and 2 mM DTT

CYP450 content: 55.0-75.0 picomoles per mg protein
Cyrotchrome b5 content: 200-300 picomoles per mg protein

P450 Activity: One unit will oxidize 1.0 picomole of dibenzylfluorescein per minute at pH 7.4 at 37 °C.
Reductase Activity: One unit will reduce 1 nanomole of cytochrome c per minute in the presence of NADPH at pH 7.8 at 25 °C.
