Rat thyroid
This thyroid cell line has been established from normal thyroid glands from 5 to 6-week-old Fischer rats. FRTL-5 is a clonal line of the continuously cultured FRTL cells derived by successive clonal isolation. The cells are characterised by maintaining highly differentiated thyroid features such as secretion of thyroglobulin and concentration of iodide. FRTL-5 cells have been used as assays measuring the stimulatory activity of human autoimmune immunoglobulins from patients with Graves disease. Tend to grow in three-dimensional structures, monolayer not formed.
Preparation Note
Coon′s modified Ham′s F12 medium (or Kaign′s modified Ham′s F12 + 45mg/L ascorbic acid + 18mg/L Inositol) + 2mM Glutamine + 10μg/ml Insulin + 10nM Hydrocortisone + 5μg/ml Transferrin + 10ng/ml gly-his-lys acetate + 10ng/ml somatostatin + 5% Foetal Bovine Serum (FBS) + 10mU/ml TSH. As an alternative to Coon′s modified Ham′s F12, it is possible to use Kaign′s modified Ham′s F12 supplemented with 45mg/L Ascorbic acid and 18mg/L Inositol.
Split sub-confluent cultures (70-80%) 1:2 to 1:3 i.e. seeding at 3-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
