General description

LTB4 protein in the sample competes with a fixed amount of alkaline phosphatase-labeled conjugate for the limited number of binding sites on the anti-rabbit LTB4 antibody. The antigen-antibody complex binds to the anti-rabbit IgG antibody-coated multiwell plate. The bound enzyme reacts with the substrate and absorbance is read at 405 nm.The intensity of the bound yellow color read at 405 nm is inversely proportional to the concentration of leukotriene in the standards or samples.

Biochem/physiol Actions

Leukotriene B4 stimulates leukocyte functions, lysosomal enzyme release, adhesion and aggregation of polymorphonuclear leukocytes and acts as a mediator of inflammatory diseases and immunoregulation.

Features and Benefits

Specifically measures LTB4 concentrations in cell culture supernatants, urine and other biological fluids with the sensitivity of 19.5 pg/mL. Negligible cross-reactivity with 20-25 related eicosanoid compounds tested (from <0.1 to 2%). Assay offers precoated plates to eliminate the coating step, all incubations proceed at room temperature, reagents are color coded and results are available in 4 hours.
Performance characteristics include low intra- and inter-assay variability and recovery from 99-111%.
