Flow Cytometry Analysis: Staining of one million mouse bone marrow cells was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1050, Anti-Mouse TER-119 (TER-119) ColorWheel^® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel^® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Rat IgG2b isotype control.

TER-119 (also known as Ly-76) is reported to be a 52 kDa protein associated with glycophorin A (GPA). It is expressed by erythroid cells from the early proerythroblast stage to mature erythrocytes and considered as a reliable erythroid lineage marker. However, its exact molecular nature has not been reported. The TER-119 epitope is not detected on hematopoietic stem cells, lymphoid cells, myeloid cells, or erythroleukemia lines. In fetal mice about 30-40% of day 10 yolk sac cells and 80-90% of day 14 liver cells display positive staining with TER-119. TER-119 positive cells in fetal liver decline after day 14 and only about 40-50% of liver cells from newborn mice are TER-119 positive. Adult mice show TER-119 expression only in about 20 to 25% of bone marrow cells and expression is not observed in thymocytes and lymph node cells. Treatment of bone marrow cells with a hypotonic shock reduces the number of TER-119 positive cells but does not affect other lineage marker positive cells. Clone TER-119 is reported to be a highly inflammatory erythrocyte-specific antibody that can induce plasma chemokines CCL2, CCL5, CXCL9, CXCL10, and CCL11 with corresponding alterations in monocyte percentages in the blood and liver within 24 hours following treatment. It is also shown to attenuate chemokine production from the synovial fluid and prevents the accumulation of inflammatory cells and complement components in the synovium. (Ref.: Crow, AR., et al. Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
