General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.
Signal transducer CD24 (UniProt: P24807; also known as Lymphocyte antigen 52, Ly-52, M1/69-J11D heat stable antigen,HSA, Nectadrin, R13-Ag, X62 heat stable antigen, CD24) is encoded by the Cd24 (also known as Cd24a, Ly-52) gene (Gene ID: 12484) in murine species. CD24 is a GPI-anchor protein without any intracellular signaling domain. It plays a pivotal role in differentiation of different cell types. Its expression is reported on hematopoietic cells, including B cells, T cells, neutrophils, dendritic cells, eosinophils, and macrophages. It is also expressed in several non-hematopoetic cells, including neuronal cells, keratinocytes, pancreatic cells, and several types of cancer cells. Its expression is shown to be higher in progenitor cells and metabolically active cells and to a lesser extent in terminally differentiated cells. In association with SIGLEC10, CD24 is reported to be involved in the selective suppression of the immune response to danger-associated molecular patterns (DAMPs), such as the high mobility group box protein 1 )HMGB1), HSP70, and HSP90. CD24 also plays a role in T cell survival. In the absence of CD24, homeostatic proliferation of T cells is markedly reduced, however immune-driven proliferation is less affected. Immunogen

Spleen cells from B 10 mice enriched for T lymphocytes and spleen cells treated with 1 pg/ml concanavalin.

Application

Quality Control Testing:

Evaluated by Flow Cytometry in C57Bl/6 mouse splenocytes.

Staining of one million mouse splenocytes was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1054, Anti-Mouse CD24 (M1/69) ColorWheel® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Rat IgG2b isotype control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Biochem/physiol Actions

Clone M1/69 is a rat monoclonal antibody that detects murine CD24.

Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Preparation Note

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
