General description

ColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.
Lymphocyte antigen 6A-2/6E-1 (UniProt: P05533; also known as Ly-6A.2/Ly-6E.1, Stem cell antigen 1, T-cell-activating protein, TAP, SCA-1) is encoded by the Ly6a (also known as Ly6) gene (Gene ID: 110454) in murine species. SCA-1 is a widely expressed GPI-anchor protein that is synthesized with a signal peptide (aa 1-26) and a propeptide (aa 113-134) that are subsequently cleaved off to produce the mature form (aa 27-112). It is expressed on pluripotent hemopoietic stem cells, some developing thymocytes, and a majority of T cells. Ly6 gene encodes for a group of antigenic specificities found primarily on T and B lymphocytes and may control various distinct alloantigens that are designated Ly-6A, B, C, D, and E. The Ly6A.2 alloantigenic specificity is found on a majority of resting spleen and lymph node cells and all activated lymphocytes. However, 6E.1 is more prominent only on activated T and B cells. Sca-1 is one the most common markers used to enrich adult murine hematopoietic stem cells (HSCs). Its expression is regulated in a complex manner in hematopoietic stem cells. When HSCs differentiate into common myeloid progenitors or lymphoid progenitors its expression is shown to be downregulated. Immature thymocytes are reported to turn off Sca-1 expression but re-express it on mature single-positive medullary thymocytes and peripheral T cells. On activated lymphocytes, its expression is upregulated. Clone D7 recognizes both Ly6A.2 and Ly6E.1 indicating a close relationship of these two forms of Ly6 gene products. Quality Control Testing

Evaluated by Flow Cytometry in mouse splenocytes.

Flow Cytometry Analysis (FC): Staining of one million mouse splenocytes was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1075, Anti-Mouse SCA-1 (D7) ColorWheel® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Rat IgG2a isotype control.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Clone D7 is a rat monoclonal antibody that detects Sca-1 (Lymphocyte antigen 6A-2/6E-1).

Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Preparation Note

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
