Flow Cytometry Analysis: Staining of one million mouse splenocytes was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1107, Anti-Mouse CD4 (GK1.5) ColorWheel^® Dye-Ready mAb and Cat. No. CWDS-PE ColorWheel^® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of Cat. No. CWA-1012 Rat IgG2b isotype control and Cat. No. CWDS-PE ColorWheel^® Antibody-Ready Phycoerythrin (PE) Dye.

T-cell surface glycoprotein CD4 (UniProt: P06332; also known as T-cell differentiation antigen L3T4, T-cell surface antigen T4/Leu-3, CD4) is encoded by the Cd4 gene (Gene ID: 12504) in murine species. CD4 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-26), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 27-394), a transmembrane domain (aa 395-417), and a cytoplasmic domain (aa 418-457). It is reported to form disulfide-linked homodimers at the cell surface. It is highly expressed in T-helper cells and its expression is also observed in macrophages, dendritic cells, and NK cells. It plays an essential role in the immune response and serves multiple functions in response to external and internal offenses. In T-cells, it functions primarily as a coreceptor for MHC class II molecule:peptide complex. It interacts simultaneously with the T-cell receptor (TCR) and the MHC class II presented by antigen presenting cells (APCs). CD4 interactions with MHC class II molecules are essential for CD4+ T cell development, activation, and effector functions. CD4 is reported to enhance the recruitment of Lck to the immunological synapse and deliver Lck to the TCR-pMHC complex. In this process the cytoplasmic tail of CD4 interacts with the N-terminal region of Lck to form a heterodimer. (Ref.: Parrish, HL., et al. (2015). PLoS One. 10(7); e0132333; Rossy, J., et al. (2012). Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
